SET8, a methyltransferase, specifically mono-methylates histone H4 at lysine 20 (H4K20), a process crucial for chromatin structure and gene expression. SET8, included in the kit, transfers a methyl group from S-adenosylmethionine (SAM) to the H4K20 peptide, generating S-adenosylhomocysteine. This is then rapidly converted to urate and hydrogen peroxide (H2O2) using the provided enzyme mix. The produced H2O2 reacts with ADHP to create resorufin, a highly fluorescent compound (excitation 530-540 nm; emission 585-595 nm).
